Stem Cell Related Patent Number US5824304
Title: | Peripheralization of hematopoietic stem cells | Inventors: | Papayannopoulou, Thalia; Seattle, WA, USA | Summary: | Described herein is a chemotherapeutic and radiotherapeutic method of treatment combined with the restoration of hematopoietic stem cells enriched from peripheral blood after treatment with the anti-VLA-4 antibody. The invention relates to methods for peripheralizing CD34+ cells, including hematopoietic stem cells, according to a process by which an anti-VLA-4 antibody is administered to a patient in order to block the binding of the VLA-4 antigen on the surface of CD34+ cells to VCAM or fibronectin, after which the peripheral blood containing the CD34+ cells is collected by leukapheresis, after which the CD34+ cells are enriched by immunoadsorption using anti-CD34 antibodies, during which time chemotherapy and/or radiotherapy are administered to the patient, after which the enriched CD34+ cells are returned to the patient's circulating blood. The invention constitutes a novel approach in the treatment of cancer and acquired immune deficiency syndrome by gene therapy. | Abstract: | The invention provides methods for peripheralizing CD34.sup.+ cells, including hematopoietic stem cells. In a first aspect, the method comprises the step of administering a blocking agent of VLA-4 antigen on the surface of CD34.sup.+ cells. In a second aspect, the method comprises administering a blocking agent of VLA-4 antigen on the surface of CD34.sup.+ cells and administering a stimulating agent of CD34.sup.+ cell proliferation in vivo. The method according to the invention is useful in the treatment of cancer or AIDS, and in gene therapy. | US Patent Website: | Click Here for Full Text of Patent | Title Number: | US5824304 | Application Number: | US1995000463298 | Date Filed: | 05/06/1995 | Date Published: | 20/10/1998 | Assignee: | None |
